METCALFE, CHARLES Area of Expertise: Kratom

We have been analyzing Kratom raw materials and finished products for over 2 years. Initially we ran samples by conventional HPLC with a run time of 30 minutes. We found this run time to be unacceptable due to the number of samples we were required to assay. Our new methodology utilized UHPLC with a reduced run time of less than 8 minutes. All of the relevant alkaloids are separated. Althought the 7-OH mitragynine is of the most interest to regulatory agencies, we have yet to find any of this isomer in either raw materials or finished products. A sample chromatogram of our new assay is attached.

PHILLIPS, TOM Area of Expertise: Various Methodologies

I have over 20 years Mass Spectrometry experience; ten of which are with hyphenated techniques, i.e., ms/ms, bot GC and LC. I have over 4 years of experience with herbal products, both the raw material and the finished products. I have developed in-house method for the determination of actives in herbals, but I do not have experience with these particular ones. I have done Folin C in several matrices for about 5 years or so.

RIMMER, CATHERINE Area of Expertise: Various Methodologies

My experience is with the methodologies (especially chromatography, absorption detection, fluorescence detection and mass spectrometric detection) and the statistics utilized for quantitation and qualitative identity testing. As the dietary supplement program coordinator at NIST, I keep up on dietary supplement matricies, analytes, and methods.

SAYRE, CASEY Area of Expertise: instrumentation and botanicals; method development & validation

I have experience with some of the instruments and techniques that could be used to detect or determine the ingredients in the call for experts. These include HPLC, mass spectrometry, and UV spectrophotometry. Additionally, I have experience with botanical sample extraction and preparation for quantitative analysis. I also have experience in the development and validation of stereospecific quantitative methods of detection of plant secondary metabolites in aglycone and glycoside forms. Most of the analytes I have experience with are chiral polyphenols.
