• Mass spectrometry common technique applied in recent years • ESI most frequently used technique for analysis and structural determination of phospholipids • ESI is sensitive with ionization occurring at site of polar head group or phosphate group • Both positive and negative modes for detection

• Ionization dependent upon length and degree of saturation of fatty acid side chain although this is effect is minor compared to the differences in head group • Source of phospholipid standards impacts upon quantitation • Analysis of phospholipids speciation can be performed by multiple reaction monitoring • Quantitative analysis of phospholipids by class can achieved by either neutral loss or precursor ion experiments
