INSTRUCTIONS TO COLLABORATORS
1. Introduction
Thank you for accepting the invitation to participate in the AOAC collaborative study for the validation of the method to quantitate gluten in incurred processed corn snack, negative rice flour, oat flour spiked with gluten from wheat, rye, and barley, blank and naturally contaminated groats, oat flakes, oat flours, and cereals. It is important to remember that this is a test of the methodology involved and not a test of the individual laboratories or their personnel. In this regard, it is very important to follow these directions and the enclosed method exactly. If for any reason you are not able to complete the study, contact the Study Director as soon as possible.
Process Sequence
Your participation will consist of three parts.
a) The first will be acknowledgment of receipt of the collaborative study package. There are many items that are being provided. Please check content of package and confirm receipt, so that you are equipped to perform the study. Please also ensure that you have enough extraction vials and ethanol p. A., which are not provided with the collaborative study package . b) The second step will be analysis of three control samples and the test kit buffer. In this step, you will be taking the test samples throughout the entire method from extraction to measurement through ELISA assay. This stage will allow you to gain familiarity with the entire sequence in the method. After sending the raw data of your analysis to the study director , the results of this step will be approved by the study director before entering the third phase . The study director may allow a re‐testing of these samples in case of questionable results. c) The third phase will consist of the analysis of the actual test materials that will comprise your portion of the study. Due to the high number of 42 samples this part is divided into 2 runs of 21 samples each. If samples are measured with higher OD values than standard 6, please re‐ measure these samples with an additional dilution. Please make sure to use the buffers stated below for further dilutions. After the analysis is complete, the results will be returned to the Study Director for review.
