Purpose or Objective To compare the disease response and loco regional control of Two Fractionation Schedules used for delivering Intracavitary Brachy therapy doses in the Treatment of Ca. Cervix and to standardize the fractionation Schedule and to Quantify the treatment outcome for the Two Fractionation Schedules e.g. Nine Gray in Two and Seven Gray in Three Fractions for the patients in locally advanced carcinoma cervix using FDG 18 PET CT Parameters.

PO-301 Superior antitumor efficacy of IFN loaded hydrogels combined with radiation against gastric cancer Q. Liu 1 , D. Zhang 1 , H. Qian 1 , B. Liu 1 1 Nanjing Drum Tower Hospital, Comprehensive Cancer Center, Nanjing, China Purpose or Objective Cytokine-induced killer (CIK) cells are often compromised by their lack of tumor targeting and long-term activity against solid tumors. Herein, we report an interferon-α (IFN-α) encapsulated hydrogel and low dose irradiation (LDI) as CIK sensitizers for gastric cancer therapy. Material and Methods IFN-α2b was incorporated in an ultraviolet-reactive, rapidly cross-linkable hydroxypropyl cellulose hydrogels. Meanwhile, LDI was used as an attractant for CIKs targeting. The bioactivity and release profiles of IFN- α2b in hydrogel were studied. The antitumor efficacy and mechanisms of IFN-α2b incorporated hydrogels. low dose irradiation and CIKs against gastric cancer were examined during in vitro and in vivo studies. Results The bioactivity of IFN-α2b was well-maintained in the hydrogels with constant release profile. In vitro studies demonstrated IFN-α2b had a synergistic effect on the cytotoxicity of CIK cells through up-regulating the expression of CD69 marker and inducing IFN-γ production. In viv o real-time image showed our hydrogels kept higher amount of drug delivery at the tumor site for a long time compared with free drug injection. Meanwhile, combination of Gel-IFN with CIK cells and LDI exhibited higher efficacy to eradicate xenograted tumor with less proliferating cells and more necrotic tumors compared with IFN-α2b or CIK cells alone. Under low dose irradiation, CIK cells could realize deep tumor This innovative combination of CIK cells, IFN-α2b loaded hydrogels with radiation cancer serve as a potent strategy for gastric cancer treatment. Radiobiology: Radiobiology of stem cells (cancer and normal tissue) PO-302 The biological role of NEAT1 long non-coding RNA in radioresistant of TNBC stem cells H.L. Chen 1 , Y.C. Lee 1 , Y.H. According to the molecular classifications of breast cancers, triple negative breast cancers (TNBCs) have a highly aggressive nature with a disproportionate number of metastatic disease cases and breast cancer deaths. There is no effective targeting therapy for TNBCs. Cancer stem cells (CSCs) are a subpopulation of cancer cells involving in tumor initiation, resistance to treatment and metastasis. Long non-coding RNAs (lncRNAs) are RNA with a length larger than 200 bp. However, the role of lncRNAs in the maintenance and the radioresistant feature of breast CSCs remain unclear. Nuclear paraspeckle infiltration. Conclusion

Material and Methods •

patients with carcinoma cervix with stage II & III will given concurrent chemo radiotherapy cisplain 40 mg/m2 weekly e.g. Chemotherapy

• • • • •

EBRT dose of 50Gy/25 Fractions Intracavitary Brachytherapy

7Gy given weekly in 3 sittings …control arm 9Gy given weekly in 2 sittings …..study arm

PET CT Parameters •

PET-CT has become an increasingly important non-invasive diagnostic tool for staging, radiotherapy planning and detection of recurrence disease in cervical cancer. Also the FDG PET derived markers SUVmax, TLG and MTV are predictive for treatment response .

Results

Conclusion PET CT should be done after two months of completion of the CRT. Patients were assessed for loco regional control and treatment response or relapse with a PET CT Quantitatively with metabolic markers and quantify the results. Advantage of PET CT evaluation is that there are no chances of missing the lymph node involvement in the para and distance metastasis as is the case with clinical evaluation. In carcinoma Cervix CRT followed by HDR ICRT with 9Gy weekly in two-fraction is equally effective in local control as 7 Gy in three fractions. Though a Pre- Treatment PET CT is recommended for proper Staging and as useful prognostic factors in cervical cancer patients.
