AOAC Official Method 2012.10. Simultaneous Determination of Vitamins E and A in Infant Formula and Adult Nutritionals
Analytes: Retinyl palmitate (cis and trans), retinyl acetate (cis and trans), -tocopherol, - tocopherol acetate
Standard: Retinyl palmitate, retinyl acetate, -tocopherol, -tocopherol acetate
Prep: Enzymatic hydrolysis and solvent extraction
Normal-phase, NH2 column, 5 um
MP: A = n-Hexane; B = 750 mL n-hexane + 250 mL methyl t-butyl ether + 3 mL methanol
Detection: Vitamin E - fluorescence, excitation/emission, 280/310 nm. Vitamin A – UV, 325 nm
Mean recovery = see tables
Limitations: 5 um column, 50 ul inj, 1.5ml/min flow, not optimum FL wavelength
