Challenges

• GC Methods – not commonly applied for M. speciosa , particularly in the wide  variety of matrices encountered – High temperatures required to elute the alkaloids – restricts  parameter adjustment for resolution of any alkaloid mixture – inadequate resolution of mitragynine and speciociliatine – Reliance on mass‐specific detection – EI, ESI, and ESI MS/MS  spectra of mitragynine, speciogynine, and speciociliatine are  nearly identical – Interference of speciociliatine – would probably not be detected  by either DAD or MS detectors – Methods should account for interference – derivatization may be necessary to overcome poor chrom. res.
