AOAC OMB Final Action Recommendation (December 2019)-2016.14

2016.14 (Jan. 2019) FOS-03 MLT Report FOR ERP USE ONLY DO NOT DISTRIBUTE

- The solution should be pipetted from the center of the bottle, not from edges or bottom.

- The unused solution should never be returned to the bottle. - The bottle should be closed immediately after each use.

- Only the 2/3 of the NaOH solution in the original bottle should be used for preparing eluents. Remaining solution can be used for other applications, including preparation of the solution for post-column addition. (k) Sodium hydroxide .—Pellets for analysis. (l) Sodium chloride .—For analysis, ACS,ISO,Reag. Ph Eur. (m) Sodium azide.— (Only when using PA1 column for HPAEC-PAD). (n) D-(-)Fructose .— ≥99.0 %. (o) D-(+)-Glucose. — ≥99.5 %. (p) N,N’-diacetylchitobiose .— >90 %. (q) Mixture of highly purified sucrase, α-amylase, pullulanase and maltase .—(Fructan Assay Kit, K-FRUC, Megazyme International Ireland Ltd, or equivalent). (r) Fructanase, mixture of highly purified recombinant exo- and endo-inulinases and recombinant endo-levanase .—(Fructan Assay Kit, K-FRUC, Megazyme International Ireland Ltd, or equivalent). Preparation of Reagents (a) Sodium hydroxide (1 M) .—Dissolve 40 ± 1 g sodium hydroxide pellets in 500 mL of water in a 1000-mL volumetric flask. After cooling down to room temperature, make up to the mark with water and mix well. (b) Sodium maleate buffer (100 mM, pH 6.5) .—Into a beaker (>500-mL) weigh 5.8 g maleic acid and dissolve with 450 mL of water (using magnetic stirrer). Adjust the pH to 6.5 with 1 M sodium hydroxide solution D(a) . Transfer the solution to a 500-mL volumetric flask and make up to the mark with water. (c) Sodium acetate buffer (100 mM, pH 4.5) .—Into a beaker (>500-mL) containing 450 mL of water, pipette 2.9 mL glacial acetic acid. Adjust to pH 4.5 with 1 M sodium hydroxide solution D(a) . Transfer the solution to a 500-mL volumetric flask and make up to the mark with water. (d) N,N’-diacetylchitobiose internal standard solution (600 µg/mL) .—Into a 25-mL volumetric flask weigh 15 mg N,N’-diacetylchitobiose and make up to the mark with water. (e) Glucose stock solution (5 mg/mL) .—Into a 25-mL volumetric flask accurately weigh 125 mg glucose and make up to the mark with water. (f) Fructose stock solution (10 mg/mL) .—Into a 25-mL volumetric flask accurately weigh 250 mg fructose and make up to the mark with water. (g) Carrez I solution .—Dissolve 10.6 g potassium hexacyanoferrate (II) trihydrate in 100 mL water and store in a brown bottle [NB: Optional reagent].
