HMWDF, IDF or SDFP (mg/100 g)
R 1
+ R 2 - P B - P A - B 2
=
X 100
+ M 2 2
M 1
where:
R 1 = residue mass 1 from M 1 in mg. R 2 = residue mass 2 from M 2 in mg. M 1 = test portion mass 1 in g; M 2 = test portion mass 2 in g. P A = ash mass from R 1 in mg. P B = protein mass from R 2 in mg.
HMWDF (%) = HMWDF (mg/100 g)/1000 IDF (%) = IDF (mg/100 g)/1000 SDFP (%) = SDFP (mg/100 g)/1000
K. CALCULATIONS for SDFS:
SDFS (mg/100 g) = R f x (Wt-IS, mg) x (PA-SDFS)/(PA-IS) x 100/M
where: R f is the response factor (see page 17).
Wt-IS is weight in mg of internal standard (Gly) contained in 1 mL of internal standard solution pipetted into sample mixture (i.e. 100 mg). AOAC Research Institute ERP Use Only PA-SDFS is the peak area of the SDFS. PA-IS is the peak area of the internal standard (glycerol; Gly). M is the test portion mass (M 1 or M 2 ) in grams of the sample whose filtrate was concentrated and analyzed by LC.
L. CALCULATION of TDF:
TDF (%) = HMWDF (%) + SDFS (%)
NOTE: These calculations can be simplified by using the Megazyme Mega-Calc TM , downloadable from where the product appears in the Megazyme web site (www.megazyme.com).
