Summary of Method:
Vitamin B12 is extracted from the sample sodium acetate buffer in the presence of cyanide at 100 °C for 30
min. Extracts are purified and concentrated with an immunoaffinity column. Vitamin B12 is determined by
ultra-high performance liquid chromatography with UV detection at 361 nm.
Method Scope/Applicability:
This method is applicable in infant formula and adult nutritional samples.
General comments about the method:
Part 1: method set up and qualification of participants
Each participating laboratory was asked to analyze two practice samples using the method provided with
the protocol and report the results to the Study Director within the required timelines (6 weeks after
receipt). Results within a range of expected levels qualified the laboratory for the second part of the study.
Part 2: Multi-laboratory test participation
Each qualified laboratory was then asked to analyze twenty MLT samples on two different days.
Each sample was assigned to either day 1 or day 2. The results were submitted to the Study
Director for evaluation.
Method Clarity:
The MLT procedure well described, clear and easy to follow.

It is necessary the use of safety protection.
