Sample Preparation
– On a specified day, collaborators will streak each coded culture to TSA and 6
different selective/differential agar media: hektoen enteric agar (HE), xylose lysine deoxycholate agar
(XLD), bismuth sulfite agar (BS), brilliant green sulfa agar (BGS), xylose lysine tergitol agar (XLT4), and
double-modified lysine iron agar (DMLIA). All media will be supplied as dehydrated culture media by the
Study Director’s laboratory and prepared in advance by the collaborating laboratories according to the
instructions provided. Streaked plates will be incubated for 24 + 2 h at 35 + 1
o
C, in accordance with the
procedures described in FDA-BAM [3] and USDA-MLG [4].
Following incubation, a collaborating technician will pick an individual colony from each plate and
resuspend the colony in 0.5 mL PBS. The technician will then randomize and blind-code the tubes
following instructions provided by the Study Director (see Appendix III). All collaborators will use the
same randomization and blind-coding scheme. The Study Director will create the sequence of random
numbers to use for sample coding [6].
