Operator 2 – Test each of the 84 PBS resuspensions in the ANSR
Salmonella
assay following the
instructions in the section “For Use as a Confirmation Test from Colony Picks” in the ANSR
Salmonella
kit insert. There are 16 positions on the ANSR reader. Samples should be tested in
groups of 15, with the 16
th
assay position reserved for a PBS blank (0.5 mL PBS, treated as a test
sample) in each experimental run. Operator 1 is not to participate in or observe this activity.
Run 1 Samples 1-15 plus PBS blank
Run 2 Samples 16-30 plus PBS blank
Run 3 Samples 31-45 plus PBS blank
Run 4 Samples 46-60 plus PBS blank
Run 5 Samples 61-75 plus PBS blank
Run 6 Samples 76-84 plus PBS blank
Each experimental run will require approximately 30 minutes.
Record test results on the Data Recording Form (appended). Record results as “Positive” or
“Negative”. If any samples produce a result of “Invalid”, record this result, then repeat the test
using the same PBS resuspension and record the second result alongside the initial result.
Day 5 Transmit Data Recording Forms and ANSR assay raw data (.json files) to the Study Director.
