Place a PCR tube holder on the PCR cooling block. Insert one PCR tube per sample into the
421
holder and remove caps. Using a multi-channel pipette, transfer 30
µ
L sample lysate to a PCR
422
tube. Cap PCR with the flat optical cap strips provided in the kit.
423
Follow screen prompts to load samples into the cycler/detector and begin the program. At the
424
completion of the PCR and detection process, follow the screen prompts to remove samples and
425
display results.
426
The results are recorded on the rack display or from a spreadsheet printout of the results (called
427
Detail View). Negative results are indicated by the green circle with (-) symbol, positive results are
428
indicated by the red circle with (+) symbol, and indeterminate results are indicated with a yellow
429
circle with (?) symbol. A yellow circle with a (?) symbol and a red slash indicated a low signal or
430
signal error.
431
Statistical Analysis was conducted as described in the AOAC INTERNATIONAL Methods
432
Committee Guidelines for Validation of Microbiological Methods for Food and Environmental
433
Surfaces [5] using the POD approach. Additionally, a Chi Square statistical analysis was
434
performed. Unless otherwise stated, all BAX® System testing was performed with and without a
435
regrowth step (10 uL transferred to 500 uL of pre-warmed BHI broth and incubated for 3 hrs at
436
37°C) and results were identical with and without this step. The exception was the dry pet food, in
437
which a regrowth was required from both enrichment methods due to PCR inhibitory compounds
438
in the primary enrichment broths.
