Listeria monocytogenes

is of particular concern for pregnant women, the aged

16

and the infirmed. It is recommended that these concerned groups avoid handling

17

this organism. Dispose of all reagents and other contaminated materials by

18

acceptable procedures for potentially biohazardous materials.

Some reagents in

19

the kit contain 1 g/L concentrations of sodium azide. Check local regulations

20

prior to disposal. Disposal of these reagents into sinks with copper or lead

21

plumbing should be followed immediately with large quantities of water to

22

prevent potential hazards.

This kit contains products of animal origin. Certified

23

knowledge of the origin and/or sanitary state of the animals does not totally

24

guarantee the absence of transmissible pathogenic agents. It is therefore

25

recommended that these products be treated as potentially infectious and handled

26

observing the usual safety precautions (do not ingest or inhale)

D. Preparation of Test Suspension

D. (a)

Pre-enrichment-

Pre-enrich test portion using filter Stomacher

®

type bags to initiate

growth of

Listeria.

For 25 g test portions, add 225 mL of pre-warmed (18-25

o

C) LPT

broth to each test portion and homogenize thoroughly for 2 minutes. For cantaloupe

32

melons, soak entire melon in approximately 1 L of pre-warmed (18-25

o

C) LPT broth.

For 125 g test portions, add 375 mL of pre-warmed (18-25

o

C) LPT broth to each test

portion and homogenize thoroughly for 2 minutes.

D. (b)

25 g test portions/cantaloupe melons rinses

: After homogenization,incubate for 26-

36

30 hours at 30 ± 1°C.

125 g test portions

: After homogenization, incubate for 24-30 hours at 30 ± 1°C.

From the primary enrichment broth, transfer a 1 mL aliquot into 10 mL of pre-

warmed (18-25

C) LPT broth and incubate for 22-26 hours at 30± 1°C.
