Analytical Range:

2 – 4375 µg/100g

2 – 10,000 µg/100s with 2.3 g sample and subsequent dilution for samples > 4000 µg/100g

Not clear if SMPR is met

LOQ:

For total vitamin B

3

(nicotinic acid + nicotinamide) = 195 µg/100g. Each analyte is therefore

<< 200 µg/100g. It would be beneficial to see the LOQ of each vitamin rather than a total.

SMPR met

Accuracy/Recovery:

Spikes at three levels, each in triplicate were 99.5% for nicotinic acid and 99.4% for

nicotinamide.

NIST 1849a averaged 107.2 µg/g (n = 4) representing 99.1% recovery against the certified value

of 108.13 ± 9.92 µg/g (mg/kg)

SMPR met

Precision (RSD

r

):

Eleven SPIFAN matrices were evaluated for precision (n=6) with the following RSR

r

:

3.5%, 5.4%, 5.4%, 6.0%, 6.0%, 5.8%, 5.0%, 5.0%, 5.6%, 5.5% and 5.1% respectively.

SMPR of ≤5% is not met in 8 of 11 samples.

Recommendation:

METHOD IS RECOMMENDED FOR FIRST ACTION

RSD

r

for the individual vitamins should be given to help decide why the repeatability was exceeded.

Can Takadiatase enzyme be omitted in future tests?

Postscript. The LOQ of the SMPR should be revisited in light of the Codex requirements for vitamin B
