Title of Method
Determination of Theanine in Tea
AOAC Candidate Method
Number (e.g. ALN-01)
TEA-01
Applicable SMPR
2015.14
Summary:
This method utilizes ionic exchange HPLC and post-column derivitization with
ninhydrin to determine the non-proteinogenic amino acid, theanine in tea, tea extracts
and tea supplements. The method utilizes a lithium citrate buffer solution as a cation-
exchange mobile phase component and as a sample extractant. Nor-leucine is used as
an internal standard. Post-column reaction with ninhydrin at 130 deg. C is followed by
UV-Vis detection at 570nm
1. Does the applicability of
the method support the
applicability of the SMPR? If
not, please explain what is
missing.
It supports the applicability of the theanine analysis portion of the SMPR. Theanine is
the only amino acid constituent on the list of analytes for the SMPR, thus necessitating
a separate analytical method.
2. Does the analytical
technique(s) used in the
method meet the SMPR? If
not, please specify how it
differs from what is stated in
the SMPR.
Yes, it meets all of the criteria
3. Are the definitions
specified in the SMPR used
and applied appropriately in
the method? If no, please
indicate how the terms are
used.
Yes, they are appropriately used.
4. Does the method, as
written, contain all
appropriate precautions and
warnings related to the
method's reagents,
components, instrumentation,
or method steps that may be
hazardous? If no, please
suggest wording or option(s).
The method contains a section detailing the safety and other precautions regarding
reagent and instrumentation use.
