Report: SPIFAN FOS-01 Single Lab Validation (SLV) for fructan in Infant Formula and Adult
Nutritionals.
1. Introduction.
During the September session (2014) at Boca Raton, the SPIFAN FOS/GOS working group
agreed for a Single Lab Validation for the FOS (fructans) determination according to the AOAC
997.08 method. The purpose is to validate the method for infant formula and adult nutritionals.
The AOAC 997.08 method is an enzymatic, chromatographic method for the determination of
fructans in food and feed products. At the time of approval of the AOAC method (1997), a
collaborative study was performed on samples with different matrices. This method is applied on a
daily basis in our lab. The method gives freedom to apply a chromatographic technique as long as
the determination of the sugars can be done accurately and without interference. Depending on the
matrix of the sample, we decide which chromatographic technique is performed, being GC, HPLC
or HPAEC.
For dairy samples our preferred method is GC because of the complexity of the matrix.
The AOAC 997.08 method is on the list of Codex approved methods for fructan determination
(Council directive 90/496/EEC 12 Dec 2012).
A yearly proficiency test is organized by “Deutsche Lebensmittel GbR” DLA in Germany where
different labs are asked to analyze for the fructan content of a double blind commercial sample.

successfully participates from 2008 on.
2. Methodology
2.1 Samples for SLV.
A Test Materials Kit, ordered at AOAC, and provided by Covance with different products of infant
and adult nutritionals like listed in the table below. As a first step the 3 placebo samples and the 12
fortified samples were screened in duplicate for the presence of fructans.
3 fortified samples (J4G-00019, J4G-00974 and J4G-00980) and also one of the placebo samples
J4G-00013 were found to contain fructans.
The pure products Orafti HP(inulin), Orafti P95(FOS) and Nutraflora P95(FOS) are part of the Test
Materials Kit and were screened in duplicate by 2 different chromatographic techniques, being GC
and HPLC.
The setup of the recovery (spiking) scheme was done in such a way that the 3 different raw
products were used over the whole analytical range.
