Fructan = (Fi+Gi)*k2
Where k2 is a correction for the water gain during hydrolysis:
k2 = (180+162*(DP-1))/(180*DP)
DP is Degree of polymerisation
2.3 Validation
2.3.1 Precision.
All samples of the Test Materials Kit were screened for the presence of fructans. Four samples
were found to contain amounts above the LOQ of 0.03 g/100 g reconstituted product.

These samples were analyzed in duplicate, on 6 different days and by different operators. As the
method is consisting of different steps, the hydrolysis, silylation, integration were performed by
different people. The Relative Standard Deviation (repeatability conditions) (RSDr) and RSD(ir)
(intermediate precision) were calculated from the obtained data. Results are found under point 3.1
and in annex 4.
2.3.2 Accuracy.
For the accuracy, a spiking was done by the 3 different raw materials provided by the Test
Materials Kit.
Nutraflora P95 (FOS)
J4G-00010
Orafti P95 (FOS)
J4G-00011
Orafti HP (inulin)
J4G-00012
The Nutraflora and Orafti P95 are fructo-oligosaccharides with DP ranging from about 2 to 8.
Nutraflora is produced from sugar as raw material while Orafti P95 is originating from chicory roots.
The mean DP is 4.
The Orafti HP is a long chain inulin extracted from chicory with DP ranging from 2 until 60 and a
mean DP of min 23. The DP for this inulin HP (J4G-00012) was found to be 34.
The raw materials as included in the Test Materials Kit were analyzed by 2 different
chromatographic techniques being GC and HPLC. No significant differences in fructan content
between the applied techniques could be observed.
Those techniques are preferred above HPAEC as this last one needs enhanced calibrations as
