Most Probable Number (MPN) Analysis.

To determine the level of

Salmonella

spp

.

in the inoculated food test portions on the day of initiation of analysis, a 5-

2

tube MPN determination is performed by the coordinating laboratory following

3

the appropriate reference method. From each of the inoculated food batches,

4

prepare 5 test portions of 100 g, 5 test portions of 25 g and 5 test portions of 10 g.

5

To each test portion, add the appropriate reference method enrichment media to

6

make a 1:10 dilution. Follow the reference method through to confirmations. Use

7

the number of positives from the high, medium and low levels to calculate the

8

MPN for each inoculated level of each matrix. Access the MPN calculator

9

( http://www.lcfltd.com/customer/LCFMPNCalculator.exe )

to determine the MPN

values and 95% confidence intervals. Data should be entered from high level to

11

low. Report MPN as MPN/test portion.

Applicability

For the detection of

species in food and feed matrices: raw ground

beef, cooked breaded chicken, raw shrimp, bagged spinach, wet pet food, and

19

pasteurized liquid whole egg.

reference. Follow all instructions carefully. Failure to do so may lead to

27

inaccurate results.

The 3M Molecular Detection Instrument is intended for use with samples that

30

have undergone heat treatment during the assay lysis step, which is designed

31

to destroy organisms present in the sample. Samples that have not been

32

properly heat treated during the assay lysis step may be considered a potential

33

biohazard and should NOT be inserted into the 3M Molecular Detection

34

Instrument.

Store the 3M Molecular Detection Assay

Salmonella

at 2-8°C. Do not freeze.

Keep kit away from light during storage. After opening the kit, check that the

38

foil pouch is undamaged. If the pouch is damaged, do not use. After opening,

39

unused reagent tubes should always be stored in the re

sealable pouch with

40

the desiccant inside to maintain stability of the lyophilized reagents. Store

41

resealed pouches at 2-8°C for no longer than 1 month. Do not use 3M

42

Molecular Detection Assay

Salmonella

past the expiration date. Expiration

date and lot number are noted on the outside label of the box.
