General
AOAC Method Number:
ANTH-003
Method Title:
Anthocyanin Profiles by HPLC with DAD and MS
Detections
Method Author(s):
Bob Durst
Reviewer Name:
John Szpylka
Summary of Method
Different fruits listed as "authentic" were squeezed into
juices. The juices were analyzed using a Synergi Hydro-
RP 80A column using different mobile phases to allow
detection by DAD or by MS.
Method Scope / Applicability:
Thirteen fruits covering seven species were studied (V.
darrowii not covered). 25 peaks were identified covering
5 classes of anthocyanidins (does not include
pelargonidin as listed in the SMPR). Sugar additions to
the oxonium were galactoside, glucoside, and
arabinoside; and acetic acid acylated on the sugar.
General Comments About the
Method:
1. The method shows the potential to profile
anthocyanins in a variety of fruits. The method targets
known compounds using two detection technologies.
2. While the ease of sample preparation and the quality
of the chromatographic analyses is appealing, the SMPR-
listed scopes of the method were not fully covered in this
report. One matrix type was studied and no exclusivity
panel fruits were included.
3. The potential of the method is evident and should be
considered if the method is more deeply evalauted using
the SMPR requirements.
Method Clarity
A general description on the preparation of samples was
present. Details on sample handling, preparation, and
storage were not listed. This could be due to the
simplicity of collecting juice from the fruits
It must be noted the only form of material tested was the
juice collecting from squeezing. No dry product, dry
extracts, capsules, softgels, or tablets were studied.
