Vitamin C in Infant Formula and Adult Nutritionals by UPLC‐UV
Manuscript Reference
In your experience using the method, does the
method perform according to the method’s
applicability as written?
In your experience with the method, is there
any safety concerns identified while using or
regarding use of the method?
Do you have any information that supports
regarding the reproducibility of the method as
written:  If so, please specify and submit
information?
As Vitamin C is unstable and oxidized to dehydroascorbic acid,
method suggests use TCEP as a reducing agent.  Dehydroascorbic acid
which has very poor response on UV may show decrease in the peak
area of ascorbic acid after keeping for long time.
LC column with 1.7 µm particle size may create problem of
backpressure after few injections due to sample matrix.
Please attach any documentation to support
your feedback, if available.
Based on your experience with the method,
are there any recommended changes to the
AOAC First Action Method as written?
Even though after using the TCEP as a reducing agent need to check
stability of the standards and sample extract. And should be
mentioned in the method. (Watchful for the response of standards
and sample extract.)
