Repeatability was evaluated by measuring six separate preparations of the two samples
on two different days. This facilitated the determination of both intraday and interday
repeatability. In general, the percentage relative standard deviation was not more than
6%. The results all the sets of the samples can be seen inTable2.
Accuracy
The accuracy of themethodwas validated using spiked recovery over three points of the
calibration range andNISTCRM 1849a. Analysing samples and comparing the analytical
result to the known added value shows the accuracy of the method. For estimating the
recovery, the samples were spiked with biotin at three levels (low, medium and high) of
the analytical calibration range to establish an acceptable recovery range for themethod.
Three recoveries were done on each level and all recoveries were between 93 to 104%.

Certified mass fraction value of biotin in NIST CRM 1849a is 199±13µg/100g. The
certified referencematerial is analysed in duplicate on two separate days. All the results
werewithin the certified limits (196, 196, 201 and206µg/100g).
Sensitivity
Limit of Quantitation (LOQ) was calculated based on the lowest working standard and
the dilution factor. The calculated LOQmeets thebiotinSMPR criteria.
