Linearity
Instrument responsewas evaluatedwith5 calibration levels over approximately2 orders ofmagnitude.
Previously, theworking standardswere reinjected as samples to verify the systemperformance over three days of
analysis. The% deviation for all injections ranged from -5.0 to 5%of target,meetingmethodperformance
criteria (data not shown). Analyses at this time also demonstrated the linear range of themethod to be 25%
above and below the calibration curve. The quantitation range for each compound is presented in table 6.
LOQ
The biotinSMPR requiresQL’s of 0.1µg/100 g for the analysis of biotin. Themethod’s current calibration range
(1.2 to120ng/mL)was establishedbasedon the range of fortification levelswithinAbbott Nutritionproducts.
ThePMQL (practicalmethod quantitation limit) for themethod as testedhas beendetermined to be~ 2.2µg/100
gor approximately22 times higher than the target QL. The PMQL calculation assumes a sample size of 1gram.
For selectedmatrices a 2 gram sample sizemaybe used todrive the PMQL lower. Additionally, based on
instrument performance it is likely that the current instrument conditionsmay allow for the calibration curve to be
extendedby asmuch as a factor of 5.
The folic acidSMPR requiresQL’s of 0.5µg/100g for the analysis of folic acid. Themethod’s current
calibration range (1.2 to120ng/mL)was established basedon the range of fortification levelswithinAbbott
Nutrition products. ThePMQL for themethod as tested has beendetermined tobe ~ 2.0µg/100 gor
approximately4 times higher than the target QL. ThePMQL calculation assumes a sample size of 1 gram. For
selectedmatrices a 2gram sample sizemaybe used todrive the PMQL lower to achieve this target level.
Additionally, basedon instrument performance it is likely that the current instrument conditionsmay allow for the
calibration curve tobe extendedby asmuch as a factor of 5.
The pantothenic acidSMPR requiresQL’s of 50µg/100g for the analysis of pantothenic acid. Themethod’s
current calibration range (8.4 to 840 ng/mL)was establishedbased on the range of fortification levelswithin
Abbott Nutritionproducts. ThePMQL for themethod as testedhas beendetermined to be ~16µg/100 gor
approximately4 times lower than the target QLwith a 1gram sample size.
ThePMQL for the analysis of niacin for themethod as tested has been determined to be~ 26µg/100gwith a 1
gram sample size based a calibration curve of 14 to1410 ng/ml.
ThePMQL for the analysis of pyridoxine for themethod as testedhas been determined tobe~ 3.3µg/100 gwith
a 1 gram sample size based a calibration curve of 1.8 to180ng/mL.
ThePMQL for the analysis of riboflavin for themethod as tested has beendetermined to be~ 2.5µg/100gwith a
1gram sample size based a calibration curve of 35 to3500 ng/mL.
ThePMQL for the analysis of thiamine for the method as testedhas been determined tobe~ 3.8µg/100 gwith a
1gram sample size based a calibration curve of 50 to5000ng/mL.
